A New Method for the Determination of Urea in Blood
نویسنده
چکیده
In a recent number of this Journal,’ a new method for the estimat, ion of urea in urine was described. This method consists in the conversion of the urea into ammonium carbonate by means of the urease of the soy bean, and an estimation of the alkalinity before and after the conversion by means of standard acid and methyl orange. The procedure is not directly applicable to the est, imation of urea in blood serum owing to the large quantity of proteins and small amount of urea present. Hpwever, by a conversion of the urea into ammonium carbonate with the use of the enzyme, and a subsequent removal of the ammonia by means of an air current as suggested by Folin,2 satisfactory results can be obtained without a preliminary removal of the proteins, as is necessary in other methods for the determination of urea in blood. Procedure. The blood is drawn in the usual manner and allowed to stand on ice until clotting is complete. As shown below the urea content of the serum does not change after standing even for three or four days; the blood can, therefore, be kept on ice over night, if desired. Two equal portions of the serum are measured into ordinary test tubes, 1 cc. of the soy bean extract3 added to one tube, and

New Enzymatic Colorimetric Method for the Quantitative Determination of Phenylalanine in Dry-Blood Spots
Phenylketonuria (PKU) is an autosomal recessive disorder, which is characterized by severe mental retardation, microcephaly and seizures. The symptoms of this disease can be prevented if detected soon after birth. Therefore, blood Phenylalanine (Phe) measurement is essential for the early diagnosis, treatment and dietary monitoring of PKU patients. The goal of this research was to introduce a r...

A new Kinetic Spectrophotometic method for the Determination of trace amount Sulfacetamide Drug into real samples has been described based with silver nanoparticles starch-capped sensor, by sodium borohydride. The reaction is followed spectrophotometrically by measuring the decrease in the absorbance at 347.5 nm. 1.0×10-2 molL-1 silver nanoparticles starch-capped sensor, 2.0×10-3 mol L-1 sodium...
متن کاملDetermination of Copper Content of Human Blood Plasma by an Ion Selective Electrode based on a New Copper-Selectophore
A new selectophore was introduced for Cu2+ ions. Spectroscopic studies showed a selectivity of a new organic compound (L) toward copper ions and several transitional metal ions. Hence, L was utilized in designing several ion selective electrodes for these cations. In practice, Cu2+ ion selective electrode behaved Nernstian (slope of 27.95±0.3 mV decade-1) over a...
متن کاملA New Potentiometric Sensor for Determination and Screening Phenylalanine in Blood Serum Based on Molecularly Imprinted Polymer
Methods routinely utilized for detection of phenylalanine in new-born blood consist of enzymatic assays, lacking sensitivity and HPLC assays which are expensive and time-consuming to conduct. We, here, report for the first time, the construction of a phenylalanine sensitive electrode, on the basis of a selective molecularly imprinted polymer, offering sensitivity, economy and ease of use for th...
متن کاملThe Determination of Blood Urea
Most of the methods for the determination of urea in blood involve the use of the enzyme urease to hydrolyze the urea, since this reaction is specific. However, instead of crystalline urease (l), crude jack bean meal extracts are used. These extracts have been found to contain other enzymes besides urease, termed “ammonia-producing” enzymes, and their substrates, which may produce large quantit...
